John Lee crebbe nel blues in quanto il suo patrigno Will Moore gli insegnò a suonare la chitarra. La principale influenza musicale dell'infanzia di Hooker fu prima il canto spirituale da chiesa, e poi, il blues. All'età di quindici anni, John Lee fuggì di casa e andò a Memphis, lavorò come maschera in un cinema, il New Daisy Theatre. Dopo essere diventato ospite fisso nei club blues di Detroit e aver suonato in tutte le feste private, John Lee iniziò la sua carriera discografica nel 1948, riscuotendo un certo successo con il suo brano "Boogie Chillen". Ad eccezione della sua tonante chitarra elettrica, lo stile di Hooker a una corda o a due corde sembrava molto simile a quello degli artisti del Delta blues degli anni '20.
